The assay uses a pre-coated 96-well microplate format and employs the sandwich enzyme immunoassay technique, with a biotinylated anti-Sonic Hedgehog detection antibody and an HRP-avidin conjugate visualised using TMB substrate. The colour reaction is read as optical density at 450 nm on a colorimetric microplate reader, with a total assay time of approximately 4 hours 30 minutes.

The kit has a reported sensitivity of 0.061 ng/ml and an assay range of 0.156-10 ng/ml.
