Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of rat Heat Shock Protein Beta 6 (HSP20) in tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay format, with an antibody specific for HSP20 pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-HSP20 Antibody and an HRP-Avidin conjugate; bound complex is visualised using a TMB substrate that, after addition of an acidic stop solution, produces a yellow colour read at 450 nm in a microplate reader.

Reported sensitivity is <0.068 ng/ml, with a standard curve range of 0.156–10 ng/ml, and a total assay time of 4 h 30 m. Reported spike-and-recovery values are 83–99% (average 91%) for serum (n=5), 80–99% (average 89%) for EDTA plasma (n=5) and 82–99% (average 90%) for heparin plasma (n=5). Reported dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is 87–109%, 74–104%, 90–94% and 86–108% for serum; 86–101%, 90–101%, 78–110% and 84–96% for EDTA plasma; and 90–101%, 77–107%, 93–96% and 89–109% for heparin plasma. Components are stored at +4 °C or −20 °C as instructed, and the kit should not be used past its expiration date.
