A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rat Heat Shock Protein 90kDa Alpha B1 (HSP90 beta) in serum, plasma, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips); HSP90 beta in standards and samples is captured by the immobilised antibody, then detected with a biotinylated anti-HSP90 beta antibody and an HRP-avidin conjugate. The reaction is visualised using TMB substrate, producing a blue product that turns yellow on addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.

Reported performance includes a sensitivity of <0.56 ng/ml and an assay range of 1.56-100 ng/ml, with a total assay time of 4h 30m. Spike-and-recovery testing (n=5) gave averages of 91% (range 80%-102%) in serum, 91% (range 81%-101%) in EDTA plasma, and 84% (range 80%-89%) in heparin plasma. Linearity was demonstrated across sample dilutions of 1:2, 1:4, 1:8 and 1:16 in all three matrices. Kit components are supplied for storage at +4°C or -20°C as instructed.
