Rat Heat Shock 70kDa Protein 5 ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of rat GRP78 BiP (Heat Shock 70kDa Protein 5) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated microplate arranged as 12 x 8 well strips. Standards and test samples are loaded into the wells and any GRP78 BiP present is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-GRP78 BiP Antibody is added and binds the captured target, then an HRP-Avidin conjugate is introduced. TMB substrate is used to visualise the reaction and the absorbance is read at 450 nm on a colourmetric microplate reader, with the concentration of GRP78 BiP calculated against the standard curve.

Performance figures supplied with the kit include a measuring range of 0.781-50 ng/ml and a sensitivity of <0.27 ng/ml, with a total assay time of 4h 30m. Spike-and-recovery testing gave average recoveries of 91% for serum (n=5, range 80%-102%), 90% for EDTA plasma (n=5, range 81%-100%) and 84% for heparin plasma (n=5, range 80%-89%), and linearity of dilution was demonstrated across 1:2, 1:4, 1:8 and 1:16 for each sample matrix. Kit components include lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4°C or -20°C as instructed.
