Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) configured for the in vitro quantitative determination of rat Heat Shock 70kDa Protein 1A (HSP70) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit is reactive with rat samples and is intended for research use in the Sandwich ELISA application.

The assay uses a pre-coated 96-well microplate (12 x 8 well strips). An immobilised antibody specific for HSP70 captures the analyte from standards and test samples; a Biotinylated Anti-HSP70 Antibody is then bound to the captured target, followed by an HRP-Avidin conjugate. TMB substrate is added and the enzymatic reaction is stopped with an acidic stop solution, producing a yellow colour whose intensity is proportional to the amount of HSP70 present. The reaction is read as absorbance at 450 nm on a colorimetric microplate reader. The standard curve covers a product range of 1.56–100 ng/ml with a reported sensitivity of 0.54 ng/ml, and the full assay is completed in 4 h 30 m.

Spike-and-recovery has been characterised for serum (range 83–100%, average 91%), EDTA plasma (90–98%, 94%) and heparin plasma (89–102%, 95%). Linearity has been examined at 1:2, 1:4, 1:8 and 1:16 dilutions across these same sample types. Kit reagents include the pre-coated microplate, lyophilised standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers.
