Rat Growth Differentiation Factor 11 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rat GDF11 in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a 96-well microplate pre-coated with an antibody specific for GDF11. Standards and samples are added to the wells, where GDF11 binds to the immobilised capture antibody. After incubation and washing, a biotinylated anti-GDF11 antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of GDF11 captured, and the concentration is calculated by reading the absorbance at 450 nm in a microplate reader against the standard curve.

The kit has a sensitivity of <5.8 pg/ml and an assay range of 15.6-1000 pg/ml, with a total assay time of 4 hours 30 minutes. Detection is colorimetric on a pre-coated microplate supplied as 12 x 8 well strips. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Reported recovery values were: serum (n=5) 83-100% with an average of 91%; EDTA plasma (n=5) 90-103% with an average of 96%; and heparin plasma (n=5) 89-102% with an average of 95%. Linearity was assessed across dilutions of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. The kit is supplied with a pre-coated 96-well microplate, lyophilised standard, Detection Solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers.
