The assay uses a pre-coated 96-well microplate and follows a sandwich enzyme immunoassay format, employing a biotinylated anti-GADD45A antibody, an HRP-avidin conjugate and TMB substrate. The enzymatic reaction yields a blue product that converts to yellow upon addition of the stop solution, and absorbance is read at 450 nm on a colorimetric microplate reader.

The reported assay range is 0.156-10 ng/ml, with a sensitivity of 0.062 ng/ml, and the total assay time is 4h 30m. Recovery values at n=5 are as follows: serum (range 83%-100%, average 91%), EDTA plasma (range 90%-100%, average 95%) and heparin plasma (range 91%-101%, average 96%).
