Rat Glycogen Synthase Kinase 3 Alpha ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of rat Glycogen Synthase Kinase 3 Alpha (GSK3 alpha) in tissue homogenates, cell lysates and other biological fluids. The assay runs on a pre-coated 96-well microplate using the sandwich enzyme immunoassay technique: standards and test samples are added to wells pre-coated with an antibody specific for GSK3 alpha, followed by incubation with a Biotinylated Anti-GSK3 alpha Antibody and then HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow on addition of acidic stop solution; the yellow intensity is proportional to GSK3 alpha concentration, calculated from the O.D. absorbance at 450 nm against a standard curve.

The colourimetric, quantitative assay has a working range of 0.625-40 ng/ml, a sensitivity of <0.227 ng/ml, and a total assay time of 4h 30m. The kit is supplied with a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers. Components should be stored at +4°C or -20°C as indicated, and the kit must not be used past its expiration date.

Sample validation was carried out using serum, EDTA plasma and heparin plasma (n=5), and linearity of dilution was demonstrated at 1:2, 1:4, 1:8 and 1:16 sample dilutions.
