Rat Glycine Dehydrogenase ELISA Kit

From Antibodies.com, this Rat Glycine Dehydrogenase ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) designed for the in vitro quantitative determination of rat Glycine Dehydrogenase in tissue homogenates, cell lysates and other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) with an immobilised antibody specific for Glycine Dehydrogenase. Standards and samples are added to the wells; after incubation and washing, a Biotinylated Anti-Glycine Dehydrogenase Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and concentration is calculated from the standard curve.

Sensitivity is <0.053 ng/ml and the assay range is 0.156–10 ng/ml. Total assay time is 4h 30m. Average recovery in serum (n=5) was 91% (range 80–102%), in EDTA plasma (n=5) 90% (range 81–100%) and in heparin plasma (n=5) 84% (range 80–89%). Linearity across dilutions 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.

Kit components include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
