Rat Glutathione Synthetase ELISA Kit

From Antibodies.com, this sandwich ELISA kit is intended for the in vitro quantitative determination of rat Glutathione Synthetase in serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated microplate bearing an antibody specific for Glutathione Synthetase. Standards and samples are added to the wells, and after washing the captured analyte is detected using a Biotinylated Anti-Glutathione Synthetase Antibody followed by an HRP-Avidin conjugate. Colourimetric detection is achieved with TMB substrate, which produces a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm on a microplate reader and the concentration of Glutathione Synthetase is calculated from a standard curve.

Reported performance figures include a product range of 0.781–50 ng/ml and a sensitivity of 0.33 ng/ml. Total assay time is 4h 30m. Kit components are stored at +4°C or -20°C as instructed. The kit is described as suitable for serum, plasma, cell lysates and tissue homogenates.

Validation data are provided for several sample matrices. Average recovery (n=5) was 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 was reported as: serum (n=5) 87–91%, 87–107%, 74–101% and 92–97%; EDTA plasma (n=5) 90–105%, 84–101%, 90–101% and 79–108%; heparin plasma (n=5) 84–95%, 92–105%, 82–105% and 89–91%.
