Rat Glutamate Receptor, Ionotropic, AMPA 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com, designed for the in vitro quantitative determination of rat Glutamate Receptor 1 (AMPA Subtype) in tissue homogenates, cell lysates or other biological fluids. The assay uses the sandwich enzyme immunoassay approach: an antibody specific for the target is pre-coated onto a 96-well microplate, standards and test samples are added, and the captured target is detected using a biotinylated anti-target antibody followed by an HRP-avidin conjugate. TMB substrate is then added and catalysed by HRP to produce a blue product that turns yellow after addition of stop solution; the intensity of yellow is proportional to target concentration and is read at 450 nm on a colorimetric microplate reader.

The quantitative assay covers a product range of 15.6–1000 pg/ml with a sensitivity of <6.1 pg/ml, and runs for a total of 4 h 30 min on a pre-coated microplate platform configured as 12 x 8 well strips. Reactivity is for rat samples, and the kit is suitable for cell lysates and tissue homogenates.

Reported recovery has been characterised in serum (n=5, range 80–102%, average 91%), EDTA plasma (n=5, range 81–100%, average 90%) and heparin plasma (n=5, range 80–89%, average 84%), with dilution linearity data provided across 1:2, 1:4, 1:8 and 1:16 dilutions. Kit components include Lyophilized Standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Components should be stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
