Rat Glutamate Decarboxylase 2, Acid ELISA Kit

Designed for the quantitative in vitro measurement of rat GAD65 (Glutamate Decarboxylase 2, Acid), this sandwich ELISA kit from Antibodies.com is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids. The assay employs a 96-well microplate pre-coated with an antibody specific for GAD65. Standards and samples are added to the wells, where GAD65 binds the immobilized capture antibody. After washing, a biotinylated anti-GAD65 detection antibody is added, followed by HRP-avidin conjugate. TMB substrate produces a blue product that converts to yellow upon addition of acidic stop solution, with absorbance read at 450 nm.

Sensitivity is 0.05 ng/ml with a working range of 0.156-10 ng/ml. Reported recovery averages are 91% for serum (range 80-102%, n=5), 90% for EDTA plasma (range 81-100%, n=5) and 84% for heparin plasma (range 80-89%, n=5). Dilutional linearity has been demonstrated at 1:2 to 1:16 across serum, EDTA plasma and heparin plasma. Total assay time is approximately 4 hours 30 minutes, with results read on a colorimetric microplate reader.

The kit includes a pre-coated 96-well microplate arranged as 12 x 8 well strips, lyophilized standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers.
