Rat Glutamate Cysteine Ligase, Catalytic ELISA Kit

This sandwich ELISA kit from Antibodies.com enables the in vitro quantitative determination of rat Glutamate Cysteine Ligase, Catalytic (GCLC) in serum, plasma, tissue homogenates, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, in which an antibody specific for GCLC is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips.

Standards and test samples are added to the wells, where GCLC is captured by the immobilized antibody. After washing, a Biotinylated Anti-GCLC Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of GCLC captured, and the concentration is calculated by reading the optical density absorbance at 450 nm in a microplate reader and referring to the standard curve.

The kit has a reported sensitivity of 0.33 ng/ml and an assay range of 0.781–50 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Recovery was reported as 80%–102% (average 91%, n=5) in serum, 81%–100% (average 90%, n=5) in EDTA plasma, and 80%–89% (average 84%, n=5) in heparin plasma.
