Rat Glucose 6 Phosphate Isomerase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Glucose 6 Phosphate Isomerase in serum, plasma or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for Glucose 6 Phosphate Isomerase that has been pre-coated onto a microplate. Standards and samples are added to the wells and the target analyte is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Glucose 6 Phosphate Isomerase Antibody is added, followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised using TMB substrate solution, producing a blue colour that changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of Glucose 6 Phosphate Isomerase captured in each well, and the concentration is calculated from the O.D. absorbance at 450 nm in a microplate reader by reference to a standard curve.

The assay has a reported range of 15.6–1000 pg/ml and a sensitivity of <5.9 pg/ml, with a total assay time of 4 hours 30 minutes. Validation data for serum and plasma samples (n=5) reported average recoveries of 80%–102% (mean 91%) for serum, 81%–100% (mean 90%) for EDTA plasma and 80%–89% (mean 84%) for heparin plasma. Linearity dilution experiments at 1:2, 1:4, 1:8 and 1:16 yielded recoveries generally within 74%–108% across the three matrices. Kit components are stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
