Rat Glucagon ELISA Kit

From Antibodies.com, this assay is a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) designed for the in vitro quantitative determination of rat Glucagon. It is intended for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and is read on a colorimetric microplate reader.

The kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to wells coated with antibody specific for Glucagon, followed by a Biotinylated Anti-Glucagon Antibody and an HRP-Avidin conjugate. A TMB substrate produces a colour change proportional to the amount of Glucagon captured, which is read at 450 nm and quantified against the standard curve. The stated product range is 23.4-1500 pg/ml with a sensitivity of <10.8 pg/ml, and the total assay time is 4 h 30 min. Included components are the pre-coated plate, lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers.

Reported spike-and-recovery values were 80-102% in serum (n=5), 81-100% in EDTA plasma (n=5) and 80-89% in heparin plasma (n=5). Dilution linearity across 1:2 to 1:16 dilutions ranged from 74-107% in serum, 79-108% in EDTA plasma and 82-105% in heparin plasma. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
