Rat Forkhead Box Protein O1 ELISA Kit

Sandwich ELISA designed for the in vitro quantitative determination of rat FOXO1A in tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate: an antibody specific for FOXO1A is immobilised on the plate, standards and samples are added and captured, and bound analyte is detected using a Biotinylated Anti-FOXO1A Antibody followed by an HRP-Avidin conjugate. TMB substrate is used to visualise the reaction, producing a blue product that turns yellow upon addition of the acidic stop solution; the intensity of yellow at 450 nm is proportional to the FOXO1A concentration in each well.

The standard curve covers 0.312-20 ng/ml, with a sensitivity of 0.136 ng/ml, and the full protocol is completed in approximately 4 hours 30 minutes. Recovery has been validated in serum (range 80%-102%, average 91%), EDTA plasma (range 81%-100%, average 90%) and heparin plasma (range 80%-89%, average 84%) across five samples per matrix.
