Rat Fibroblast Growth Factor 1, Acidic ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of rat Fibroblast Growth Factor 1 (FGF1), also referred to as acidic FGF, in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for FGF1 pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, followed by a biotinylated anti-FGF1 antibody and HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution; the density of yellow is proportional to the FGF1 captured in each well and is read by O.D. absorbance at 450 nm on a colorimetric microplate reader.

The standard curve covers a product range of 31.2–2000 pg/ml, with a reported sensitivity of 11.4 pg/ml. Sample recovery values provided are: serum (n=5) 80%–102% (average 91%); EDTA plasma (n=5) 81%–100% (average 90%); and heparin plasma (n=5) 80%–89% (average 84%). Dilution linearity data is included for serum, EDTA plasma, and heparin plasma at 1:2, 1:4, 1:8, and 1:16 dilutions.

The kit is supplied with a pre-coated 96 well microplate configured as 12 x 8 well strips, lyophilized standard, detection solution A, detection solution B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution, and plate sealers. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date. Total assay time is 4h 30m.
