From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Mitochondrial Ferritin in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) bearing an antibody specific for Mitochondrial Ferritin. Standards and test samples are added to the wells and the target present in each sample is captured by the immobilised antibody. Following a wash step, a Biotinylated Anti-Mitochondrial Ferritin Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Mitochondrial Ferritin captured in each well, and concentration is calculated by reading the O.D. absorbance at 450 nm against the standard curve.

The kit shows a sensitivity of <0.113 ng/ml and an assay range of 0.312-20 ng/ml. Total assay time is 4h 30m, with colorimetric detection on a pre-coated microplate platform. Recovery has been demonstrated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (range 81%-100%, average 90%, n=5), and heparin plasma (range 80%-89%, average 84%, n=5). Dilution linearity across 1:2 to 1:16 has been reported for serum (74-107%), EDTA plasma (79-108%), and heparin plasma (82-105%). Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
