From Antibodies.com, a sandwich Enzyme-Linked Immunosorbent Assay (ELISA) enables the in vitro quantitative determination of rat Fatty Acid Transport Protein 1 (FATP1) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for FATP1, a biotinylated anti-FATP1 detection antibody, and an HRP-avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, with the resulting colour intensity measured at 450 nm and proportional to the FATP1 concentration in each sample.
Quantitative performance is reported with a sensitivity of <0.071 ng/ml across a standard range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Reported recovery ranges are 80%–102% in serum (average 91%, n=5), 81%–100% in EDTA plasma (average 90%, n=5), and 80%–89% in heparin plasma (average 84%, n=5). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 is reported in the corresponding ranges for serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%), each with n=5.
Components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
