From Antibodies.com, this Sandwich ELISA is designed for the in vitro quantitative determination of rat Farnesoid X Receptor (also known as Bile Acid Receptor NR1H4) in tissue homogenates, cell lysates, and other biological fluids. The assay uses the sandwich enzyme immunoassay technique, employing an antibody specific for Bile Acid Receptor NR1H4 pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by Biotinylated Anti-Bile Acid Receptor NR1H4 Antibody and HRP-Avidin conjugate. A TMB substrate solution is used to visualise the reaction, producing a blue product that changes to yellow upon addition of acidic stop solution; the concentration of target is calculated by reading O.D. absorbance at 450nm in a colorimetric microplate reader and referring to the standard curve.

The standard range is 0.156–10 ng/ml with a sensitivity of <0.061 ng/ml, and the total assay time is 4h 30m. Reported recovery values include 80–102% (serum, average 91%), 81–100% (EDTA plasma, average 90%), and 80–89% (heparin plasma, average 84%), each from n=5. Dilution linearity data are provided at 1:2, 1:4, 1:8, and 1:16 for serum, EDTA plasma, and heparin plasma. The kit is supplied with a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard, Detection Solution A and Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, with components stored at +4°C or -20°C as applicable.

Reactivity is rat-specific, and the kit is suitable for cell lysates and tissue homogenates. Components should not be used past the stated expiration date.
