A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com for the in vitro quantitative determination of rat Fas Ligand in serum, plasma, tissue homogenates or other biological fluids. The assay employs a pre-coated 96-well microplate with an antibody specific for Fas Ligand immobilised on the wells. Standards and test samples are added; Fas Ligand present is bound by the immobilised antibody. After washing, a Biotinylated Anti-Fas Ligand Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the concentration of Fas Ligand is calculated from the standard curve.

The quantitative assay has a reported assay time of 4h 30m, a sensitivity of <0.076 ng/ml and an assay range of 0.312–20 ng/ml. The manufacturer source lists values at n=5 for serum (80–102% / 91%), EDTA plasma (81–100% / 90%) and heparin plasma (80–89% / 84%). Per-dilution values across 1:2, 1:4, 1:8 and 1:16 sample dilutions are also reported for the same three sample types (n=5).

Components supplied, per the manufacturer source, include a pre-coated microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at +4°C or −20°C as instructed in the source, and the kit should not be used past its expiration date.
