Rat Epidermal Growth Factor Receptor 2 ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of rat Epidermal Growth Factor Receptor 2 (ErbB2/HER2) in serum, plasma or other biological fluids. The kit employs a pre-coated 96-well microplate format and is intended for use with a colorimetric microplate reader.

The assay uses the sandwich enzyme immunoassay technique. Standards and test samples are added to wells pre-coated with an antibody specific for ErbB2/HER2, where the analyte is captured. After washing, a biotinylated anti-ErbB2/HER2 antibody binds the captured analyte, followed by addition of an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction; the resulting absorbance at 450 nm is proportional to the amount of ErbB2/HER2 present and is quantified against a standard curve.

Reported performance includes an assay range of 0.625–40 ng/ml and a sensitivity of 0.221 ng/ml, with a total assay time of 4h 30m. Recovery values are 80–102% in serum (n=5, average 91%), 81–100% in EDTA plasma (n=5, average 90%), and 80–89% in heparin plasma (n=5, average 84%). Components include a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with storage at +4°C or –20°C as instructed.
