Rat Elastase 2, Neutrophil ELISA Kit

A sandwich ELISA kit supplied by Antibodies.com for the in vitro quantitative determination of Neutrophil Elastase in rat serum, plasma, tissue homogenates and other biological fluids. The assay is run on a pre-coated 96-well microplate arranged as 12 x 8 well strips and uses a biotinylated anti-Neutrophil Elastase detection antibody with HRP-Avidin conjugate and TMB substrate; the enzymatic reaction is visualised by a blue-to-yellow colour change and quantified by reading the absorbance at 450 nm on a colorimetric microplate reader.

Performance specifications include a product range of 0.156–10 ng/ml and a sensitivity of <0.069 ng/ml, with a total assay time of 4h 30m. Reported spike-and-recovery values are 91% average (range 80%–102%, n=5) for serum, 90% average (81%–100%, n=5) for EDTA plasma, and 84% average (80%–89%, n=5) for heparin plasma. Dilution linearity across 1:2, 1:4, 1:8 and 1:16 gave recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.

Kit contents comprise a pre-coated 96-well microplate (12 x 8 well strips) stored at –20°C, lyophilized standard (2 vials) at –20°C, Detection Solution A (120 µl) at –20°C, Detection Solution B (120 µl) at –20°C, 30X Wash Buffer (20 ml) at +4°C, Sample Dilution Buffer (45 ml) at –20°C, TMB Substrate (9 ml) at +4°C, Stop Solution (6 ml) at +4°C and 5 adhesive plate sealers, with components stored at +4°C or –20°C as instructed.
