Rat Dopamine Receptor D4 ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat dopamine receptor D4 (DRD4) in tissue homogenates, cell lysates, or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for DRD4 is pre-coated onto a 96-well microplate (12 x 8 well strips), and standards or test samples are added so that DRD4 present in each sample binds to the immobilised antibody. After washing, a Biotinylated Anti-DRD4 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of DRD4 captured per well, and the concentration is calculated from the absorbance at 450 nm read against a standard curve on a colorimetric microplate reader.

The stated assay range is 0.156–10 ng/ml with a sensitivity of <0.057 ng/ml, and the assay is completed in approximately 4 hours 30 minutes. Recovery has been evaluated: serum samples (n=5) gave 80%–102% (average 91%), EDTA plasma (n=5) gave 81%–100% (average 90%), and heparin plasma (n=5) gave 80%–89% (average 84%). Dilution linearity has been tested at 1:2, 1:4, 1:8, and 1:16 across serum, EDTA plasma, and heparin plasma. Kit components should be stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date.
