The Antibodies.com Rat Dab1 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) for the in vitro quantitative determination of rat Disabled Homolog 1 (Dab1) in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips. An antibody specific for Dab1 is immobilised on the plate; standards and samples are added and any Dab1 present is captured, then detected using a Biotinylated Anti-Dab1 Antibody and an HRP-Avidin conjugate. The reaction is visualised with TMB substrate and the absorbance read at 450 nm on a colorimetric microplate reader.

According to the manufacturer, the assay has a sensitivity of 0.107 ng/ml and a standard range of 0.312–20 ng/ml, with a total run time of 4 hours 30 minutes. Reported recovery (n=5) averages 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Dilution linearity has been assessed at 1:2, 1:4, 1:8 and 1:16 across the same matrices, with serum values of 87–91%, 87–107%, 74–101% and 92–97%, EDTA plasma values of 90–105%, 84–101%, 90–101% and 79–108%, and heparin plasma values of 84–95%, 92–105%, 82–105% and 89–91%.

The kit is supplied with a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
