Rat Dipeptidyl Peptidase IV ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of rat Dipeptidyl Peptidase IV (DPP4) in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique: an antibody specific for DPP4 is pre-coated onto a microplate, standards and test samples are added to the wells, and DPP4 present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-DPP4 Antibody binds the captured target and an HRP-Avidin conjugate is added. Following incubation and washing, TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of an acidic stop solution. The density of yellow is proportional to the amount of DPP4 captured in each well, and the concentration is calculated by reading the absorbance at 450nm in a microplate reader against the standard curve.

The assay has a quantitative range of 62.5-4000 pg/ml and a sensitivity of <26 pg/ml, with a total run time of 4h 30m. Reported spike-and-recovery averages from the manufacturer are 91% in serum, 90% in EDTA plasma and 84% in heparin plasma. Parallel dilution-linearity testing across 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries ranging from 74% to 108% across serum, EDTA plasma and heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
