Rat Diacylglycerol-O-Acyltransferase Homolog 1 ELISA Kit

A sandwich-format enzyme-linked immunosorbent assay (ELISA) supplied under the Antibodies.com brand enables in vitro quantitative determination of rat Diacylglycerol O Acyltransferase Homolog 1 (DGAT1) in serum, plasma, tissue homogenates and other biological fluids.

The procedure employs a pre-coated microplate onto which an antibody specific for DGAT1 has been immobilised. Standards and test samples are dispensed into the wells, allowing DGAT1 to be captured by the coated antibody. After incubation and washing, a biotinylated anti-DGAT1 antibody is added, followed by an HRP-avidin conjugate and further incubation. TMB substrate solution is then applied; HRP-catalysed conversion produces a blue product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader, and DGAT1 concentration is calculated by reference to the standard curve.

Manufacturer-reported characteristics include a sensitivity of <0.33 ng/ml and an assay range of 0.781-50 ng/ml, with the full protocol completed in approximately 4 hours 30 minutes. Spike-and-recovery performance has been evaluated in serum (range 80% - 102%, average 91%), EDTA plasma (range 81% - 100%, average 90%) and heparin plasma (range 80% - 89%, average 84%).
