Rat Desmin ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of rat Desmin in serum, plasma, tissue homogenates and other biological fluids. The kit is supplied as a pre-coated 96-well microplate arranged as 12 × 8 well strips, with an antibody specific for Desmin immobilised on the plate surface. Standards and test samples are added to the wells and any Desmin present binds to the capture antibody; after incubation and washing, a biotinylated anti-Desmin detection antibody is added, followed by an HRP conjugate and TMB substrate solution. The enzymatic reaction produces a blue colour that turns yellow after addition of acidic stop solution, and absorbance is read at 450 nm on a colorimetric microplate reader. Desmin concentration is calculated by reference to the standard curve.

Reported assay sensitivity is <0.117 ng/ml with an assay range of 0.312–20 ng/ml in one configuration, and <0.188 ng/ml with an assay range of 0.313–20 ng/ml in another. Recovery in neat serum has been reported at 84–100% (average 92%, n=5) and 89–105% (average 96%, n=10) respectively; EDTA plasma recovery is 83–99% (average 91%, n=5) and 90–104% (average 97%, n=10); heparin plasma recovery is 85–101% (average 93%, n=5) and 86–103% (average 93%, n=10). The assay is specific for Desmin with no reported cross-reactivity with other analogs or variants. Serum and plasma samples should be diluted at least 1:2 with the supplied sample dilution buffer prior to testing, and kit components should be stored at +4 °C or −20 °C as instructed in the protocol manual. The kit should not be used past its expiration date.
