Rat Deiodinase, Iodothyronine, Type I ELISA Kit

Manufactured by Antibodies.com, this Sandwich ELISA is intended for the quantitative in vitro determination of rat DIO1 (Deiodinase, Iodothyronine, Type I) in tissue homogenates and other biological fluids. The assay uses a pre-coated microplate in a sandwich enzyme immunoassay format, in which standards or samples are added to immobilised anti-DIO1 wells, followed by a biotinylated anti-DIO1 detection antibody and an HRP-avidin conjugate. Signal is generated with TMB substrate, terminated with acidic stop solution, and read at 450 nm, with the concentration of DIO1 calculated against a standard curve.

The quantitative range is 0.156–10 ng/ml with a reported sensitivity of <0.059 ng/ml, and the protocol can be completed in approximately 4 hours 30 minutes. Reported recoveries (n=5) are serum, average 91% (range 80–102%); EDTA plasma, average 90% (range 81–100%); and heparin plasma, average 84% (range 80–89%). Dilution linearity across 1:2 to 1:16 for serum, EDTA plasma and heparin plasma falls within 74–108%. Reagents are stored at +4°C or –20°C as indicated, and the kit should not be used past its expiration date.
