Rat Cytochrome P450 1A1 ELISA Kit

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is designed for the in vitro quantitative determination of rat Cytochrome P450 1A1 (CYP1A1) in tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is intended for use with rat samples and is read on a colorimetric microplate reader at 450 nm, with absorbance proportional to CYP1A1 concentration as determined against a standard curve.

The standard curve range is 0.312–20 ng/ml with a sensitivity of <0.129 ng/ml. The assay is performed on a pre-coated microplate and has a total run time of 4 hours 30 minutes. Kit components include a pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The principle involves a CYP1A1-specific capture antibody, a biotinylated anti-CYP1A1 detection antibody and an HRP-avidin conjugate, with signal developed by TMB substrate and acidic stop solution.

Validated recovery has been reported across sample matrices: serum 80–102% with an average of 91% (n=5); EDTA plasma 81–101% averaging 91% (n=5); and heparin plasma 80–89% averaging 84% (n=5). Dilution linearity has also been assessed at 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma samples.
