From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of rat Cytochrome C (Somatic) in tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with colorimetric detection by O.D. absorbance measured at 450 nm in a microplate reader. Total assay time is 4 hours 30 minutes, and kit components are stored at +4°C or -20°C as instructed.
Quantitative performance is supported by a standard curve range of 1.56–100 ng/ml and a sensitivity of <0.58 ng/ml. The protocol uses a biotinylated anti-Cytochrome C detection antibody with an HRP-Avidin conjugate and TMB substrate, producing a blue product that turns yellow after addition of acidic stop solution; the resulting colour intensity is proportional to the amount of Cytochrome C captured in each well.
