Rat Cystathionine Gamma Lyase ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com, designed for the in vitro quantitative determination of rat Cystathionase/CTH (Cystathionine Gamma Lyase) in tissue homogenates or other biological fluids.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for Cystathionase/CTH is pre-coated onto a microtiter plate. Standards and test samples are added into the wells, where the target is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-Cystathionase/CTH Antibody is added and binds to the captured target. Following further washing, an HRP-Avidin conjugate is added. TMB substrate solution is then used to visualise the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Cystathionase/CTH captured in each well. Concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to the standard curve.

The kit has a sensitivity of <0.053 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery data: serum (n=5) range 80-102%, average 91%; EDTA plasma (n=5) range 81-100%, average 90%; heparin plasma (n=5) range 80-89%, average 84%. Dilution linearity at 1:2, 1:4, 1:8, 1:16: serum 87-91%, 87-107%, 74-101%, 92-97%; EDTA plasma 90-105%, 84-101%, 90-101%, 79-108%; heparin plasma 84-95%, 92-105%, 82-105%, 89-91%. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The product is supplied ready-to-use.
