Rat Cyclin Dependent Kinase Inhibitor 1A ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of rat p21 (Cyclin Dependent Kinase Inhibitor 1A) in tissue homogenates, cell lysates or other biological fluids. The assay is run on a pre-coated 96-well microplate and read on a colorimetric microplate reader at 450 nm.

The method uses an antibody specific for p21 that is immobilized on the plate. Standards and samples are added to the wells and any p21 present is captured. After washing, a Biotinylated Anti-p21 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then added; catalysis produces a blue product that turns yellow upon addition of acidic stop solution, and the yellow colour intensity is proportional to the amount of p21 captured. The p21 concentration is calculated by reference to a standard curve.

Reported performance includes an assay range of 0.156-10 ng/ml and a sensitivity of 0.068 ng/ml, with a total assay time of 4h 30m. Mean recovery was 91% in serum (n=5, range 83-100%), 94% in EDTA plasma (n=5, range 90-98%) and 95% in heparin plasma (n=5, range 89-102%). Kit components include a pre-coated microplate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with storage at +4°C or -20°C as indicated.
