Rat Cyclin Dependent Kinase 5 ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of rat Cyclin Dependent Kinase 5 (CDK5) in tissue homogenates, cell lysates and other biological fluids. An antibody specific for CDK5 is pre-coated onto a microplate. Standards and samples are added to the wells, where CDK5 is captured by the immobilised antibody, then detected using a biotinylated anti-CDK5 antibody and HRP-avidin conjugate. The reaction is visualised with TMB substrate, producing a yellow product after the addition of acidic stop solution whose absorbance is read at 450 nm and compared to a standard curve.

Reported sensitivity is <0.059 ng/ml and the assay range is 0.156–10 ng/ml. In validation work (n=5), recovery averaged 91% (range 80%–102%) in serum, 90% (range 81%–100%) in EDTA plasma and 84% (range 80%–89%) in heparin plasma. Across linear dilution steps of 1:2, 1:4, 1:8 and 1:16, recovery ranges were 87%–91%, 87%–107%, 74%–101% and 92%–97% for serum (n=5); 90%–105%, 84%–101%, 90%–101% and 79%–108% for EDTA plasma (n=5); and 84%–95%, 92%–105%, 82%–105% and 89%–91% for heparin plasma (n=5).

Total assay time is 4h 30m and the detection method is colorimetric, read on a microplate reader. Kit components should be stored at +4°C or −20°C as instructed and not used past the expiration date.
