Description

This Antibodies.com kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of rat CD38, referred to in the product listing as cyclic ADP-ribose hydrolase, in serum, plasma, tissue homogenates or other biological fluids. The assay uses a pre-coated microplate in a 12 x 8 well strip format, with detection performed by reading the optical density at 450 nm in a colorimetric microplate reader.

Sensitivity is reported as <1.24 ng/ml, with a quantitative working range of 3.12–200 ng/ml and a total assay time of approximately 4 hours 30 minutes. In the procedure, standards and samples are added to wells pre-coated with an antibody specific for CD38, followed by incubation with a biotinylated anti-CD38 detection antibody and an HRP-avidin conjugate. TMB substrate is then added to produce a blue colour that turns yellow upon addition of the acidic stop solution; the intensity of yellow is proportional to the amount of CD38 captured in each well, and concentration is calculated against the standard curve read at 450 nm. Standards are prepared by a 1:2 serial dilution series from a 200 ng/ml stock, with the lowest standard at 3.13 ng/ml and the sample dilution buffer serving as the zero standard.

Supplied components include the pre-coated 96-well microplate (12 x 8 well strips), two vials of lyophilized standard, detection solution A (120 μl), detection solution B (120 μl), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml), and adhesive plate sealers. Kit components should be stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.
