A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Cross Linked C-Telopeptide Of Type II Collagen in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The kit is supplied as a pre-coated 96-well microplate (12 x 8 well strips). Standards and samples are added to the wells, followed by a Biotinylated Anti-Cross Linked C-Telopeptide Of Type II Collagen Antibody and an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow on addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration is calculated against the standard curve.

The standard product range is 125-8000 pg/ml with a sensitivity of 33.5 pg/ml, and the total assay time is 4 h 30 min. Recovery across n=5 samples was reported as 80-102% (average 91%) for serum, 81-100% (average 90%) for EDTA plasma, and 80-89% (average 84%) for heparin plasma. Linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma (n=5 for each matrix).

Kit components include a pre-coated 96 well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and five adhesive plate sealers. Components are stored at -20°C or +4°C as instructed, and the kit should not be used past its expiration date.
