A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of rat Corticosteroid Binding Globulin (also referred to as Cortisol Binding Globulin) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips and is read on a colorimetric microplate reader at 450 nm.

The product range is 3.12–200 ng/ml with a sensitivity of <1.39 ng/ml, and the total assay time is 4h 30m. The principle is a standard sandwich enzyme immunoassay: a capture antibody specific for Cortisol Binding Globulin is immobilised on the plate; standards and samples are added and the target binds to the wells; after washing, a biotinylated anti-Cortisol Binding Globulin detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution; the absorbance at 450 nm is proportional to the amount of target captured.

Validated recovery ranges for rat samples (n=5) are: serum 80% – 102% (average 91%), EDTA plasma 81% – 100% (average 90%) and heparin plasma 80% – 89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was: serum 87–91%, 87–107%, 74–101% and 92–97%; EDTA plasma 90–105%, 84–101%, 90–101% and 79–108%; heparin plasma 84–95%, 92–105%, 82–105% and 89–91%. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
