Description

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com, intended for the in vitro quantitative determination of rat Coronin 1a/TACO in tissue homogenates, cell lysates, and other biological fluids. The kit is reactive with rat samples and employs a pre-coated 96-well microplate (12 x 8 well strips) bearing an antibody specific for Coronin 1a/TACO immobilized on the wells.

Standards and test samples are pipetted into the wells, where target analyte is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-Coronin 1a/TACO detection antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then applied to visualise the enzymatic reaction, yielding a blue-coloured product that changes to yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the concentration of Coronin 1a/TACO is calculated by reference to the standard curve. The reported assay sensitivity is <0.128 ng/ml with a measuring range of 0.312-20 ng/ml, and the total assay time is approximately 4 hours 30 minutes.

Reported recovery values from the manufacturer include rat serum (range 83-100%, average 91%, n=5), EDTA plasma (range 90-98%, average 94%, n=5), and heparin plasma (range 89-102%, average 95%, n=5). Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
