Rat Complement Component 3 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of rat Complement C3 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. A 96-well microplate is pre-coated with an antibody specific for Complement C3. Standards and samples are added to the wells; the captured Complement C3 is detected using a Biotinylated Anti-Complement C3 Antibody followed by an HRP-Avidin conjugate. TMB substrate generates a blue product that becomes yellow after addition of acidic stop solution, and absorbance is read at 450 nm in a microplate reader with concentration calculated from the standard curve.

Reported sensitivity is <10.62 ng/ml, the product range is 31.2-2000 ng/ml, and the total assay time is 4 hours 30 minutes. Spike-and-recovery (n=5) gave ranges and averages of 80%-102% (91%), 81%-99% (90%) and 80%-89% (84%) for serum, EDTA plasma and heparin plasma, respectively. Linearity at dilutions of 1:2, 1:4, 1:8 and 1:16 (n=5) was 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Detection is colorimetric on a pre-coated 96-well microplate (12 x 8-well strips). Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past the expiration date.
