Rat Collagen Type XIX ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com is intended for the in vitro quantitative determination of rat COL19A1 in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a biotinylated anti-COL19A1 detection antibody and an HRP-avidin conjugate added in sequence. The HRP enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that changes to yellow after addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the concentration of COL19A1 calculated against the standard curve.

Quoted performance characteristics include a measuring range of 0.312–20 ng/ml and a sensitivity of <0.116 ng/ml. The total assay time is 4h 30m. Spike-and-recovery data across five replicates gave average recoveries of 91% for serum (83%–100%), 96% for EDTA plasma (90%–103%) and 95% for heparin plasma (89%–102%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries of 83–110%, 79–100%, 90–98% and 82–108% for serum (n=5); 90–98%, 90–105%, 73–109% and 87–91% for EDTA plasma (n=5); and 88–103%, 77–105%, 95–98% and 87–110% for heparin plasma (n=5).

Supplied components include the pre-coated microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, with individual storage temperatures specified between +4°C and -20°C.
