Rat Collagen Type I Alpha 2 ELISA Kit

From Antibodies.com, this Sandwich ELISA is designed for the in vitro quantitative determination of rat Collagen Type I Alpha 2 (COL1A2) in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for COL1A2 pre-coated onto a microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-COL1A2 Antibody and an HRP-Avidin conjugate. A TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of the stop solution. The absorbance is read at 450 nm in a microplate reader and the concentration of COL1A2 in each sample is calculated against the standard curve.

The assay has a stated sensitivity of <0.55 ng/ml and an assay range of 1.56–100 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Detection is colourimetric and read on a standard microplate reader. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Validated sample types include serum, EDTA plasma and heparin plasma. Reported recovery ranges are 80–102% (serum, average 91%), 81–101% (EDTA plasma, average 91%) and 80–89% (heparin plasma, average 84%), each evaluated with n=5. Dilution linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions, with reported ranges across dilutions of 70–107% for serum, 81–110% for EDTA plasma and 77–107% for heparin plasma (n=5 per sample type).
