Rat Cofilin 1, Non Muscle ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of rat Cofilin in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, where an antibody specific for Cofilin has been pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells and the Cofilin present is bound by the immobilized antibody. After incubation and washing, Biotinylated Anti-Cofilin Antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the Cofilin concentration, which is calculated by reading O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

Sensitivity is 0.059 ng/ml and the assay range is 0.156-10 ng/ml. Assay time is 4h 30m and the platform is a pre-coated microplate of 12 x 8 well strips. Recovery in serum (n=5) ranges from 80-102% with an average of 91%; EDTA plasma (n=5) ranges from 81-100% with an average of 90%; heparin plasma (n=5) ranges from 80-89% with an average of 84%. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 dilutions, with serum (n=5) ranging 74-107%, EDTA plasma (n=5) ranging 79-108% and heparin plasma (n=5) ranging 82-105% across dilutions. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date. Detection is colourimetric, read on a microplate reader.
