The assay uses an antibody specific for Coagulation Factor V pre-coated onto a microtiter plate. Standards and test samples are added, and captured target is detected using a biotinylated anti-Coagulation Factor V antibody followed by an HRP-Avidin conjugate. TMB substrate is then added; catalysis produces a blue colour that turns yellow upon addition of acidic stop solution, with absorbance read at 450nm on a colorimetric microplate reader.
