Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit is designed for the in vitro quantitative determination of rat CD163 in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique in which an antibody specific for CD163 is pre-coated onto a 96-well microplate. Standards and test samples are added to the wells so that CD163 is captured by the immobilised antibody. After washing, a Biotinylated Anti-CD163 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction; the blue product turns yellow upon addition of acidic stop solution, and the absorbance is read at 450 nm on a colorimetric microplate reader.

According to the manufacturer, the assay has a sensitivity of <0.113 ng/ml and a measuring range of 0.312–20 ng/ml. The total assay time is 4 h 30 min. The platform is a pre-coated microplate formatted as 12 × 8 well strips, and kit components are stored at +4°C or -20°C as instructed; components should not be used past the expiration date.

Validated sample types include serum, EDTA plasma and heparin plasma. Reported average recovery values are 91% for serum (range 83–100%, n=5), 94% for EDTA plasma (range 90–99%, n=5) and 96% for heparin plasma (range 91–101%, n=5). Dilution linearity for serum (n=5) at 1:2, 1:4, 1:8 and 1:16 is reported as 81–99%, 90–102%, 77–104% and 88–95% respectively, with parallel ranges for EDTA plasma (94–104%, 80–108%, 88–93%, 86–108%) and heparin plasma (76–100%, 92–99%, 88–105%, 85–99%).
