Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of rat Cluster Of Differentiation 14 (CD14) in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using a 96-well microtitre plate pre-coated with an antibody specific for CD14. Standards and test samples are added to the wells, where any CD14 present is captured by the immobilised antibody. After washing, a Biotinylated Anti-CD14 Antibody is added, followed by an HRP-Avidin conjugate and a TMB substrate solution. The enzymatic reaction produces a blue colour that changes to yellow upon addition of the acidic stop solution; the density of yellow is proportional to the amount of CD14 captured, and is measured as O.D. absorbance at 450 nm on a colourmetric microplate reader.

Quantitative performance characteristics stated by the manufacturer include an assay range of 1.56-100 ng/ml and a sensitivity of <0.51 ng/ml. Reported average recoveries are 91% for serum, 96% for EDTA plasma, and 95% for heparin plasma (n=5 per sample type). Manufacturer-supplied dilution data cover 1:2 to 1:16 preparations across each of these matrices. The total assay time is 4h 30m, and kit components should be stored at +4 °C or –20 °C as instructed and not used past the expiry date.
