Manufactured by Antibodies.com, this sandwich enzyme-linked immunosorbent assay (ELISA) kit is designed for the in vitro quantitative determination of rat Clara Cell Protein 16 (referred to in the manufacturer documentation as Uteroglobin) in serum, plasma, tissue homogenates, cell lysates, lung lavage fluid, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique: an antibody specific for Uteroglobin is pre-coated onto a microtiter plate, standards and test samples are added, and Uteroglobin in each sample is captured by the immobilized antibody. After washing, Biotinylated Anti-Uteroglobin Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Uteroglobin captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a reported product range of 78.1–5000 pg/ml and a sensitivity of 35 pg/ml, with a total assay time of 4h 30m. Reported average recovery values are 91% for serum (range 83%–100%, n=5), 96% for EDTA plasma (90%–103%, n=5) and 95% for Heparin plasma (89%–102%, n=5). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 is reported as: Serum (n=5) 83–110%, 79–100%, 90–98%, 82–108%; EDTA Plasma (n=5) 90–98%, 90–105%, 73–109%, 87–91%; Heparin Plasma (n=5) 88–103%, 77–105%, 95–98%, 87–110%. The kit is intended for use with a colorimetric microplate reader, and components should be stored at +4°C or –20°C as instructed.
