The Antibodies.com Rat Cholecystokinin ELISA Kit is a competitive enzyme-linked immunosorbent assay (cELISA) for the in vitro quantitative determination of rat cholecystokinin in serum, plasma or other biological fluids. The assay employs a pre-coated microplate onto which cholecystokinin present in samples or standards competes with immobilized antigen for binding sites on a biotinylated anti-cholecystokinin antibody. After incubation and washing, an HRP-avidin conjugate is added. A TMB substrate solution then visualises the enzymatic reaction, producing a blue colour that turns yellow on addition of an acidic stop solution. The density of yellow is inversely proportional to the amount of cholecystokinin present, and the concentration is calculated from the O.D. absorbance read at 450 nm on a colourimetric microplate reader.

The assay has a detection range of 12.5–800 pg/ml and a sensitivity of 3.75 pg/ml, with a total assay time of 2 h 30 min. Reported average recoveries are 91% for serum (range 80%–102%, n=5), 90% for EDTA plasma (range 81%–100%, n=5), and 84% for heparin plasma (range 80%–89%, n=5). Dilution linearity (1:2 to 1:16, n=5) has been reported as 87–107% for serum, 84–108% for EDTA plasma, and 82–105% for heparin plasma. Kit components should be stored at +4°C or −20°C as instructed and the product must not be used past its expiration date.
