Rat Cathepsin L ELISA Kit

From Antibodies.com, this Sandwich ELISA kit is designed for the in vitro quantitative determination of rat Cathepsin L in serum, plasma, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for Cathepsin L that is pre-coated onto a microplate. Standards and test samples are added to the wells, where Cathepsin L is captured by the immobilised antibody, followed by incubation with a Biotinylated Anti-Cathepsin L/MEP Antibody and an HRP-Avidin conjugate. A TMB substrate solution produces a blue product that turns yellow upon addition of an acidic stop solution; the yellow intensity is proportional to the Cathepsin L concentration, read at 450 nm on a microplate reader.

Detection is colorimetric and the assay has a sensitivity of 0.225 ng/ml with a measuring range of 0.625–40 ng/ml. The pre-coated microplate is supplied as 12 x 8 well strips and the total assay time is 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.

Reported average recovery values are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5), and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity data at 1:2, 1:4, 1:8, and 1:16 are also reported for serum, EDTA plasma, and heparin plasma.
