From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is designed for the in vitro quantitative determination of rat CELSR2 in tissue homogenates or other biological fluids. The assay targets Cadherin EGF LAG Seven Pass G-Type Receptor 2 with reactivity for rat samples, and runs on a colourmetric, pre-coated microplate platform (12 x 8 well strips) with an assay time of 4h 30m.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for CELSR2 is pre-coated onto a 96-well microtiter plate; standards and samples are added, followed by Biotinylated Anti-CELSR2 Antibody and then HRP-Avidin conjugate. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a microplate reader and CELSR2 concentration is calculated by reference to the standard curve.

The reported product range is 0.781–50 ng/ml with a sensitivity of <0.24 ng/ml. Recovery is reported for serum (80–102%, average 91%), EDTA plasma (81–101%, average 91%) and heparin plasma (80–89%, average 84%) across dilutions of 1:2 to 1:16. Kit components include a pre-coated 96-well microplate, lyophilised standard (2 vials), Detection Solution A and B (120 µl each), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers.
