From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) enables the in vitro quantitative determination of rat C-Peptide in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay employs a competitive enzyme immunoassay technique using a 96-well microtiter plate pre-coated with C-Peptide antigen. C-Peptide present in samples or standards competes with the immobilized antigen for binding sites on a Biotinylated Anti-C-Peptide Antibody. After washing to remove unbound antibody, an HRP-Avidin conjugate is added, followed by TMB substrate that yields a blue colour, turning yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, with colour intensity inversely proportional to the amount of C-Peptide in each sample or standard.

The kit has a measuring range of 125-8000 pg/ml and a sensitivity of 33.27 pg/ml. The platform is a pre-coated microplate of 12 x 8 well strips, with a total assay time of 2 h 30 min. Components should be stored at +4°C or -20°C as indicated, and the kit must not be used past its expiration date. Reported average recovery values are 91% for serum, 90% for EDTA plasma, and 93% for heparin plasma (n=5 each).
